I need something to lift my spirits and just make me laugh. I rented a few hopefuls this weekend and they were DUDS. all the funny parts were in the previews and the stories were stupid.

If you haven't seen Office Space, that's a must. And if you don't already watch Modern Family, start from the beginning.
My go to is always "I love Lucy"! She is one funny lady!!!! Favorite episodes are Vitameatavegamin(lucy does a tv commercial), California Here we Come, Ricky Needs an Agent, and Lucy does The Tango. The Naked Gun is hilarious and so is The Campaign-just came out this summer. Also for a quick pick me up watch Abbot and Costello's "Who's on First?"

You didn't specify movies, so I wonder if you would be interested in a comedy series? On an overseas plane trip, our TV's didn't work, and the sweet flight attendant had a season of The Big Bang Theory on her iPad and lent it to me to watch. It was great! (I love Sheldon, though I guess his roommate, Leonard is really my favorite character.) The writers for that series are just marvelous and some of the humor is terrific. The more you watch, the more you want to watch (or so it is for me!)

If you don't mind some potty humor and girls acting bad, "Bridesmaids" is super hilarious! I laughed out loud at many points during that movie. Office Space is funny, too.

"Tower Heist" with Eddie Murphy, Ben Stiller, Casey Affleck, and a ton of other great actors. A few dirty parts (verbal, not visual) but HILARIOUS!
I guess it depends on what type of comedy. There is the romantic comedy, rated r comedy and witty comedy.
For romantic and witty comedy where it kind of takes your mind somewhere else I would recommend "Midnight in Paris". If you want to laugh with a comedy that is raunchy/curse words and keeps you entertained, my thought is "Hangover 1" or "Made". I think you probably need a comedy that is unexpected. Good luck.

I really enjoyed Couples Retreat, the one with Vince Vauhgn(sp?), it was super cute. Very light hearted, touching, with lots of laughs. :)
What kind of humor do you like? Humor is a tricky thing. I for one thought Something About Mary was OK, but The Hangover left me flat. I like comedians like Gabriel Iglasias and Eddie Izzard. I thought Hot Fuzz and Shaun of the Dead were hilarious. One of my favorite movies is The Princess Bride.
I also liked What About Bob? And any of the Ghostbusters movies. Miss Congeniality is another good one.
O Brother Where Art Thou and Best in Show were two I surprisingly liked very much. And Bill and Ted are just silly fun. Back to the Future? Men In Black? A Knight's Tale?

City Island is my very favorite movie ever. It's a clever comedy with Andy Garcia and Juliana Margulies that came out in 2010. It's on Netflix. I've recommended it to tons of people, and I show it to my seniors (in high school), and I've never heard of anyone who didn't love it. :)
This Means War is a great funny movie.
Hit up YouTube and look up dumbest Family Feud answers. Or Whose Line is it Anyway clips. Those always crack me up!

Couples Retreat was a hilarious movie! For TVs shows, Duck Dynasty has really been cracking me up lately. :)
"The Dinner Game" which is the English translation of this great funny well known French comedy film.
"Le Diner de Cons." is the French title of it. (I don't know how to type out the accent over the "i" in the title on my keyboard).
Everyone has different taste so it depends on what makes you laugh and what is just plain stupid. I would think that you can't go wrong with the oldies. Can you rent seasons of The Cosby Show, I Love Lucy, Leave It to Beaver, etc.? That's what I would tape and watch when I was up all night with my babies way back then. They kept me laughing so I didn't resent my newborns' crazy schedules.

Muriel's Wedding - my all time favorite. Great humor. Its an Australian movie but you might recognize some people in it from American movies.
More of a dark comedy.. which I find funny is Harold and Maude.. I can watch this movie over and over and still find interesting things about it..
it's not for every one but I found it quite amusing.
We really laugh a lot at The Other Guys, and Date night.
If your looking for a quicker uplift, I had a cashier at the grocery store recommend the TV show impractical jokers. I have laughed out loud with the show a few times since.
An older one that always cracks me up is What About Bob? with Bill Murray and Richard Dreyfuss.

If you want an older one, Without a Paddle is so funny! It is Seth Green and Matthew Lillard and a big bear. I love that one.
A newer one that I could not believe was so incredibly funny was The Three Stooges. The new one with Sean Hayes (from Will and Grace). I actually did wet my pants, not too hard to do since kids, but still takes some effort from laughing! I have to give this 2 thumbs up and so did my curmudgeony husband who never laughs at funny movies.
I also watched Here Comes the Boom and this one was pretty funny because it was so sappy and unbelievable. But it wasn't as funny as the Three Stooges.

I like many that people have written below, but one I haven't seen mentioned is Dinner for Schmucks. I laughed through the entire thing.
I love the old film 'See No Evil, Hear No Evil' - the language is pretty filthy but the concept is HILARIOUS!! I also second America's Funniest Home Videos!!
I second Office Space and all of the Meet the Fockers. Duplex with Ben Stiller is funny too. Oh and if you haven't seen Fun with Dick and Jane it is H.I.L.A.R.I.O.U.S! It's with Jim Carrey so you know it's gotta be funny, LOL. I could do without all these comedies coming out with the crude humor and nudity.

Pollyanna. It's not a comedy and it was made a long time ago, but it always cheers my mother up. She suffers bouts of depression and it helps her.
You could also try Funny Farm.
Looks like you've got plenty of suggestions by now but I have to add: "Death at a Funeral" - either version and "I Now Pronounce You Chuck and Larry." I get movies all the time from the library since tv is the pits lately.
And like a lot of people on here, I'm also sick of the "F" word and the filthy sex scenes. It's like they're just trying to see how many times they can use the "F" word in one sentence or something. When did that become the thing to do?!!! It's getting harder and harder to find "clean" comedies!! Plus, it's actually getting harder to just find good comedies in general!
